      采用邻苯二甲醛柱前衍生5种氨基酸,用反向高效液相色谱法测定衍生化产物,通过优化衍生化条件和色谱条件,使该方法在0.25 μmol/L~25 μmol/L范围内线性良好。方法检出限为0.08 μmol/L~0.12 μmol/L,标准溶液测定6次的RSD为0.1%~1.2%,空白加标样的回收率为80.3%~115%。用该方法测定实际水样,5种氨基酸测定值均低于0.514 μmol/L,加标回收率在75.5%~120%之间。

      In this study, a precolumn derivatization method using O phthalaldehyde(OPA) followed by reversed phase high performance liquid chromatography for determination of five amino acids was developed. By optimizing the derivatization factors and chromatography factors, the good linear relationships were obtained in the range of 0.25 μmol/L~25 μmol/L. Under the optimum conditions,the detection limits were from 0.08 μmol/L to 0.12 μmol/L. The relative standard deviation by six times determination and the spiked recoveries of blank were 0.1%~1.2% and 80.3%~115%, respectively. The method was applied to the determination of free amino acids in actual surface water. The results showed that the concentration of amino acid in water were below 0.514 μmol/L and the spiked recoveries were from 75.5% to 120%.
