用超高效液相色谱-质谱法(UPLC-MS)测定经6-氨基喹啉基-N-羟基琥珀酰亚胺基氨基甲酸酯(AQC)柱前衍生后的两种神经毒素β-N-甲氨基-L-丙氨酸(BMAA)和2,4-二氨基丁酸(DAB),通过优化试验条件,使方法在1.00 μg/L~250 μg/L范围内线性良好,BMAA 和DAB 的方法检出限分别为0.5 μg/L和0.9 μg/L。对空白蓝藻样品做低、中、高3个质量浓度水平的加标回收试验,BMAA和DAB的平均回收率分别为(88.2±4.0)%和(87.7±14.2)%,6次测定结果的RSD分别为2.6%~7.9%和2.1%~11.3%。 |

Two neurotoxins,β-N-methylamino-L-alanine (BMAA)and 2,4-diaminobutyric acid(DAB) were determined by ultra-performance liquid chromatography-mass spectrometry(UPLC-MS), using pre-column derivatization with 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate (AQC). By optimizing the test conditions, the method had good linearity in the range of 1.00 μg/L~250 μg/L. The detection limits of BMAA and DAB were 0.5 μg/L and 0.9 μg/L respectively. The average spiked recoveries of BMAA and DAB at low, medium and high concentration levels in blank samples were (88.2±4.0)% and (87.7±14.2)%, respectively, and the RSDs of 6 measurements were 2.6%~7.9% and 2.1%~11.3% respectively. |
